血管内皮生长因子对生物衍生骨复合骨髓基质细胞体内成骨的影响 被引量:18

摘要 目的研究在生物衍生骨复合骨髓基质细胞治疗骨缺损过程中,血管内皮生长因子对骨折愈合及移植骨内成骨的影响。方法用青紫蓝兔30只制作右侧桡骨骨缺损模型,将预先制作好的复合骨髓基质细胞生物衍生骨植入。随机数字法分为两组,分别应用血管内皮生长因子和血管内皮生长因子抗体。在术后2,4,6,8周摄X线片及常规组织学检查观察骨折愈合及移植骨内成骨情况。结果X线显示,血管内皮生长因子组所有骨缺损植骨均成功愈合。血管内皮生长因子抗体组有一只未愈合。组织学评分显示血管内皮生长因子组形成软骨和新骨最好。与血管内皮生长因子抗体组相比,在2,4,6,8周时,t值分别为3.091,3.361,2.982,2.317,P值分别小于0.01,0.01,0.05,0.05。结论生物衍生骨复合骨髓基质细胞治疗骨缺损过程中,缺乏血管内皮生长因子会严重影响移植骨内骨的形成,阻碍骨折的愈合。应用外源性的血管内皮生长因子可通过增加骨折端的血流量促进骨折愈合。 Aim To observe the effect o vascular endothelial growth factor on the healing of fracture and osteogenesis of transplant bone in healing bone defect treated with bio derived bone compounded with marrow stroma cells (MSC).Methods Marrow stroma cells cultured from 30 Qingzilan rabbits in vitro were seeded into cancellous bone(bio derived bone) andcubated for 72 h. Radius defect of right sidedness were created by surgery in 30 Qingzilan rabbits and treated with bio derived bone. Then 30 models were randomly divided into VEGF group and VEGF antibody group by numerical method. X ray films and routine histology examination were conducted in two groups 2, 4, 6, 8 weeks after respectively.Results The X ray films showed that, except one bone defect was not healed, the others bone defects were repaired rapidly in VEGF antibody group as well as in VEGF group.The histology examination showed that cartilages and new bones in VEGF group were better than those in VEGF antibody group. In comparison with VEGF antibody group, the histological scores were 2.00±0.41 2weeks after in VEGF group (t=3.091,P< 0.01); 2.96±0.20 4 weeks in VEGF group(t=3.361, P< 0.01); 3.72±0.61 6 weeks after in VEGF group(t=2.982,P< 0.05); 4.76±0.52 8 weeks after in VEGF group(t=2.317, P< 0.05).Conclusion In the process of healing bone defects with bio derived bone, the lack of VEGF can seriously influence the form of bones and hinder the healing of fracture. Ectogenous VEGF may facilitate the healing of fracture by promoting the blood flow of fracture tip.
