纤维连接蛋白对多发性骨髓瘤化疗敏感性及其临床价值的研究

摘要 目的探讨纤维连接蛋白对多发性骨髓瘤化疗敏感性及其临床价值。方法采用MTT法测量柔红霉素、羟基喜树碱、金喜素和康莱特注射液对骨髓瘤细胞的细胞毒性 ;流式细胞仪定量检测凋亡细胞并计算细胞凋亡倍数。结果纤维连接蛋白能使骨髓瘤耐受化疗药物的细胞毒作用 ,柔红霉素、羟基喜树碱、金喜素和康莱特注射液的耐药倍数分别为 8.3、9.7、5 .2和 4 .4 ;并能抑制化疗药物诱导的细胞早期凋亡 ,上述化疗药物的细胞凋亡倍数分别为 0 .31、0 .6 0、0 .5 7和 0 .33。结论纤维连接蛋白能降低多发性骨髓瘤对化疗药物的敏感性 ,通过抑制化疗药物诱导的细胞凋亡 ,参与骨髓瘤早期耐药的形成 ,从而为临床逆转多发性骨髓瘤耐药提供新的思路。 Objective To investigate the function of fibronectin in the chemotherapy sensitivity to multiple myeloma and its value. Methods The cytotoxicity of daunoblastin, hydroxycamptothecin, topotecan and kanglaite injection to myeloma cell was measured by application of MTT. Apoptosis cell was quantitatively detected and the apoptosis times were counted with flow cytometry. Results Fibronectin decreased the sensitivity of myeloma to chemotherapeutic medications.The resistance factors for daunoblastin, hydroxycamptothecin, topotecan and kanglaite injection were 8.3, 9.7, 5.2 and 4.4, respectively, which could inhibit the early apoptosis of cells induced by chemotherapeutic medications, with the cell apoptosis times of 0.31, 0.60, 0.57 and 0.33. Conclusion Fibronectin can reduce the sensitivity of multiple myeloma to chemotherapeutic medications and is involved into the formation of early resistance of myeloma through inhibiting the cell apoptosis induced by chemotherapeutic medications, thus providing a new thought for reversing drug resistance to multiple myeloma in clinic.
