摘要 目的 探讨全反式维甲酸 (ATRA)对培养大鼠主动脉血管平滑肌细胞增生及P2 1蛋白表达水平的影响。方法 血管平滑肌细胞 (VSMC)采用组织贴块法培养。ATRA(2 .5×10 -6mol·L-1)作用于经 2 0 %胎牛血清刺激后的血管平滑肌细胞 ,2 4h后分别行 3H TdR掺入实验测定和P2 1蛋白印迹检测P2 1蛋白表达。结果 ATRA明显抑制血管平滑肌细胞DNA合成和促进P2 1蛋白表达。结论 ATRA促进P2 Objective To investigate the effect of all-trans retinoic acid(ATRA)on proliferation of rat vascular smooth muscle cells and the expression of P21. Method Vascular smooth muscle cells(VSMC) were cultured by tissue explant method, VSMC stimulated by 20% fetal bovine serum were acted with ATRA(2.5×10 -6-mol·L -1). 3H-TdR assay and P21Western-Blotting were determined respectively after 24-h.Result ATRA inhibited synthesis of DNA of vascular smooth muscle cells DNA and enhanced the expression of P21 protein in rats.Conclusion Enhanced expression of P21 with ATRA is one of reasons for inhibiting proliferation of vascular smooth muscle cells of rat.
