摘要 目的 :探讨慢速冷冻保存对人成熟卵子和未成熟卵子发育潜能的影响。方法 :在行宫腔配子移植和多囊卵巢综合征患者未成熟卵穿刺周期中收集不同成熟期的卵子 10 5个 ,其中成熟卵 (MII) 6 9个 ,未成熟卵 (MI和GV) 36个。各成熟期卵子经慢冻 速融后进行形态学和存活力观察 ,并进一步观察体外成熟、受精及胚胎培养情况。结果 :经慢速冷冻后各个成熟期卵子的形态都发生了改变。冷冻后的未成熟卵子可以体外成熟 (76 6 7% )。冻融成熟卵子受精后的分裂率 (76 92 % )高于体外成熟卵受精后的分裂率 (P <0 .0 1) ,只有冻融成熟卵子经胚胎培养获得优质胚胎 (33 33% )。结论 :用慢速冷冻法保存成熟卵子可以得到较好的胚胎发育结果 ,未成熟卵子的冷冻保存仍然存在相当的难度。 Objective:To study the effect of slow-freezing method on the developmental potential of the human mature and immature oocytes.Methods:105 oocytes were collected from the patients undergoing GIUT and PCOS treatment cycles:69 were mature oocytes(MⅡ)and 36 were immature(MI and GV).All the oocytes were cryopreserved with slow-freezing method.After thawing,survival and morphological evaluation were given to the different maturity oocytes.The thawed immature oocytes were subjected to in vitro maturation.Intracytoplasmic sperm injection(ICSI)was applied for insemination of the high quality mature oocytes and the outcomes of insemination and subsequent embryo development were also recorded.Results:The oocytes at different maturity had morphological changes after slow-freezing-rapid-thawing.The maturation rate for the frozen-thawed immature oocytes(76.67%) was similar to the rate of fresh immature oocytes.There was no significant difference for the survival rates,high-quality rates and fertilization rates between the frozen-thawed mature and immature oocytes.The cleavage rate of the zygotes from the mature oocytes(76.92%)was significantly higher than the immature oocytes( P <0.01).High-quality embryos were developed from the frozen-thawed mature oocytes.Conclusions:The mature oocytes can be well cryopreserved using slow-freezing method.There is still many problens as for cryopreservation of immature oocytes.
