人参皂甙单体Rb1联合异博定对多药耐药细胞系K562/HHT的耐药逆转作用

Reversing effects of saponins Rb1 combined with VRE on human leukemic multi-drug resistant cell line K562/HHT

目的 探讨中药钙离子通道拮抗剂人参皂甙单体Rb1联合异博定 ,对白血病多药耐药细胞系 (K 5 6 2 /HHT)的耐药逆转作用。方法 药物敏感试验采用四唑氮盐 (MTT)比色试验法。结果 2 0 μmol/LRb1、10 μmol/LRb1联合 5 μmol/L异博定耐药逆转倍数分别为 4 .4、12 .5倍。结论 Rb1联合异博定可以显著增强Rb1逆转活性。

Objective To explore the reversing effects of saponins Rb1 combined with VRE on human leukemic multi drug resistant cell line K562/HHT in vitro.Methods The cytotoxic effect was determined by MTT colorimetric assay.Results Rb1 at 0～80mol/L did not cause obvious cytotoxicity on K562/HHT. The reversal of Rb1 to K562/HHT in 20μmol/L, 40μmol/L combined with VRE was 4.4 fold, 12.5 fold respectively.Conclusion Rb1 combined with VRE can reverse multi drug resistance of K562/HHT in vitro increasely.
