反义4CL1基因转化烟草调控木质素生物合成 被引量:12

摘要 将控制木质素生物合成的关键限速酶 (香豆酸CoA连接酶 ,4CL)基因 4CL1反向插入植物表达载体pBI12 1构建表达载体 ,通过农杆菌叶盘法介导转化烟草 ,获得一批转基因抗性植株 .经PCR检测 ,证明 4CL1基因已成功反义转入到烟草植株中 .通过对转基因植株和对照植株的纤维素和木质素含量的对比分析 ,发现反义 4CL1基因转化植株的纤维素含量比对照提高了 11 4 % ,而木质素含量比对照降低了 19 1% . The anti sense 4CL1 expression vector pBIFA1 was constructed by anti inserting 4 coumarate:CoA ligase (4CL1) gene into binary vector pBI121 The construction was transferred into tobacco by Agrobacterium mediated transformation method. Regenerated tobacco plantlets were verified by PCR, which indicated that the 4CL1 gene was integrated into the genomes of transformed plantlets. The lignin content of transgenic plantlets reduced by 19 1% compared to the control, while the cellulose content increased by 11 4%.
