摘要
通过把雪花莲外源凝集素基因(GNA)与潮霉素抗性基因(Hyg)构建到同一表达载体上,利用基因枪转化方法,把雪花莲外源凝集素基因导入粳稻保持系中,通过PCR方法和潮霉素抗性筛选,检测该基因的导入情况,发现部分克隆植株只具有单一潮霉素抗性,需要结合抗虫性测定来确定GNA基因的导入及表达。
This experiment was constructed the GNA gene and marker gene(H yg )into the same ex-pression plasmid.Then it was used to transform the contain ing line of rice by microprojectile-mediat-ed transformation.The results of P CR and resistance to Hyg indicated that some plants only have one type of resis tance,only resistance to Hyg.It is required to combine the resistance to insec t to make sure that GNA gene has been transformed into rice and has expressed.
