摘要
应用 PCR- Am p- FL P技术对 75例陕西正常老年人群、94例老年动脉粥样硬化 (AS)患者与 71例陕西正常青年人群的 YNZ2 2 - VNTR(可变数目串联重复序列 variable num ber tandem repeats)位点进行多态性分析。结果显示 ,在正常青年人群中 YNZ2 2位点共检测出 2 9种基因型 ,1 0个等位基因 ,其片段大小在 1 6 8~ 938bp,杂合度为 70 .4 % ,多态信息量 (polym orphism inform ation content,PIC)为 0 .84 ;老年人群中 YNZ2 2基因点共检测出 2 0种基因型 ,9个等位基因 ,其片段大小在 1 6 8~ 798bp,杂合度为 2 4 % ,多态信息量 (PIC)为 0 .84 ,与青年群体的基因频率分布有显著差异 ;动脉粥样硬化 (AS)人群中 YNZ2 2基因点共检测出 2 3种基因型 ,9个等位基因 ,其片段大小在 1 6 8~ 72 8bp,杂合度为 2 4 .4 7% ,多态信息量为 0 .81。表明老年群体杂合度显著降低 ,最主要的是在基因型数目与大基因片段上老年群体与青年群体差异显著。 YNZ2 2位点多态性改变或不稳定可能是引起衰老的原因之一。
The polymorphism at YNZ22 VNTR locus was studied by PCR Amp FLP in 71 cases of Youth,94 cases of atherosclerosis and 75 cases of normal old people of Shaanxi.Results show that:① 29 genotypes and 10 alleles ranging from 168 bp to 938 bp were detected in the 71 normal youth subjects.② 20 genotypes,9 alleles ranging from 168bp to 798bp were revealed in normal old people of Shaanxi.The heterozygosity of YNZ22 was 24%,PIC 0.84;③ 23 genotypes,9 alleles ranging from 168 bp to 728 bp were revealed in atherosclerosis cases.The heterozygosity of YNZ22 was 0.24,PIC 0.81.④ There is obvious difference in distribution of allele frequency in two groups (Old and Youth);and the heterozygosity of old people decreases obviously.There is obvious difference in the number of genotypes and large alleles in two groups.⑤ Our results suggest that unstability of YNZ22 locus can be related to senility.
