摘要
以农艺性状较好的 2 4份小麦品种 (系 )为材料 ,筛选出了愈伤组织诱导率高且植株再生能力强的基因型材料山农 995 6 0 4和F2 81- 10 ,在此基础上对影响愈伤组织的分化能力以及较长时间保持分化能力的因素进行了研究。结果表明 ,在愈伤组织的继代过程中 ,保持较高浓度的 2 ,4 -D ,或对愈伤组织进行交替继代培养有利于较长时间保持其再生能力。最佳取材时间在开花后 15d左右 ,而对于在开花后 2 5d取的大龄胚 ,若仅挑取盾片组织进行接种 。
Genotypes with higher regeneration frequency were obtained from 24 different wheat genotypes with good agronomic traits,and the factors influencing the induction and differentiation of wheat callus were studied .The result showed that keeping high concentration 2?4-D in subculture or alternating subculture was beneficial to retain higher regeneration frequency of wheat callus.Good callus with high regeneration frequency can be obtained when scutellum of embryo at 25 days after pollination were used as explant even.
