摘要
将以两相成珠法制备的琼脂珠,经三聚氯氰活化后与对苯二胺偶联,得到对氨基苯基琼脂珠衍生物。以此作为固定化载体,经重氮化共价偶联了羊精囊的前列腺素合成酶。研究了制备固定化酶时加入酶量与固定化酶活力的关系以及固定化酶的使用稳定性和热稳定性。
The immobilization of prostaglandin synthetase from microsomes of sheep vesicular glands by means of covalent binding to carrier is described. Agar beads were activated with cyanuric chloride, then coupled with p-phenyienediamine, became p-aminophenyl agar bead which was used as carrier. After activated by diazotization, the carrier was coupled with microsomes containing prostaglandin synthetase. The relationship between the amount of added native enzyme and the activity of the immobilized enzyme; the operational stability and the thermal stability of the immobilized enzyme were studied.
