清除补体对大鼠局灶性脑缺血的作用 被引量:5

Effects of complement depletion on focal cerebral ischemia in rats

摘要 目的观察抑制补体系统的活化或消除其影响抑制多形核中性粒细胞浸润,是否能减轻局灶性脑缺血损伤程度和神经功能缺损。方法采用局灶性脑缺血大鼠模型,用眼镜蛇毒因子(cobravenomfac-tor,CVF)消耗大鼠体内补体,于缺血第7天进行形态学观察,于缺血第1,4,7天测试实验动物的神经行为功能。结果与单纯缺血组比较,CVF处理组大鼠脑前囟前1.2mm和后0.8mm、1.8mm冠状平面梗死面积占同侧半球面积的百分数减小(29±3)%/(35±4)%,(32±5)/(45±8)%,(28±4)%/(38±4)%,达到非常显著性意义(t=3.13,4.30,4.47,P<0.01);缺血区域内中性粒细胞数均比单纯缺血组减少(3.4±1.4)/(5.3±2.0),达到显著性意义(t=2.35,P<0.05);CVF处理组的神经功能损害比单纯缺血组轻,达到显著性意义(P<0.05)或非常显著性意义(P<0.01)。结论清除体内补体对局灶性脑缺血损伤有保护作用,减轻神经功能缺损程度;局灶性脑缺血过程中补体系统活化,并对脑缺血损伤有贡献。 Aim To observe whether the inhibition of activity or elimi-nation of complement system in inhib iting infiltration of polymorphic n ucleus can induce the level of focal cerebra l ischemia and neurologic impairmen t system.Methods Twenty-four Wistar rats were subjec ted to the focal cerebral ischemia models of rats and then divided into three group of eigh t rats:control group,pseudo-operat ion group and treatment group with co bra venom factor eliminating complemen t in rats(CVF group).Morphology was observed at the 7th day after ischemi a among the three groups and neural behavior function of all the rats was measured at the 1st,4th,7th day post-ischemia respectively.Results The area percent of coronal plan infa rct size in the homo-hemisphere of the CV F group reduced to(29.3±3.3)%at1.2mm before bregma,(31.7±5.0)%at 0.8mm behind bregma ,and(28.5±4.3)%at 1.8mm behind bregma respectively,significantly dif-ferent from those of the control grou p(35.2±4.1)%,(45.4±7.5)%and(38.0±4.3)%(t=3.13,4.30,4.47,P<0.01).The number of neutral cells of the GVF group reduce d to 3.4±1.4in the ischemia region,significantly lower than that of the control group(5.3±2.2)(t=2.35,P<0.05).The neurologic impairment of the CV F group was lighter or sig-nificantly lighter than that of the c ontrol group(P<0.05or P<0.01).Conclusion The depletion of complement protect s focal cerebral ischmia injury and reduces the level of neuro logic impairment.The complement ac-tivated during the course of focal cerebral ischima contributes to cereb ral ischemia injury.
