摘要
目的 观察血管生成抑制剂TNP 4 70对血管瘤血管内皮细胞体外增殖的影响 ,并与氢化可的松相比较。方法 选择一草莓状血管瘤 ,采用M199培养液按组织块法进行血管内皮细胞(VEC)体外培养。传至第 3代 ,VEC分为 6组 :组 1、2、3在培养液中加入内皮细胞生长支持物(ECGS) ,组 1为对照组 ,组 2加入氢化可的松 ,组 3加入TNP 4 70 ;组 4、5、6培养液中加入ECGS和雌二醇 (E2 ) ,组 4为对照组 ,组 5加入氢化可的松 ,组 6加入TNP 4 70。培养后 3、6、9d分别行细胞计数和 3H TdR掺入检测。结果 至第 9d ,组 1VEC增殖明显 ,约增至 3倍。组 2、组 3VEC增殖受到抑制 ,仅为组 1的 1/10 (均P <0 .0 1)。组 4VEC增殖最为显著 ,约为组 1的 2倍。组 5、组 6VEC增殖仍受到明显抑制 ,约为组 4的 1/7(均P <0 .0 1)。组 2与组 3、组 5与组 6VEC增殖状况相似 (均P>0 .0 5 )。结论 TNP 4 70能明显抑制ECGS或ECGS和E2 联合诱导的血管瘤VEC体外增殖 ,抑制作用与氢化可的松程度相仿。
Objective To test in vitro of the inhibitory effect of TNP 470 on proliferation of vascular endothelial cells (VECs) from hemangioma, and compare with hydrocortisone.Methods VECs from one specimen of strawberry hemangioma were cultured in medium 199 with endothelial cell growth supplement (ECGS). Based on different culture conditions and treatment, the VECs of 3rd generation were divided into 6 groups as: 1) control; 2) supplemented with hydrocortisone; 3) supplemented with TNP 470; 4) estradiol (E2); 5) E2 and hydrocortisone, and 6) E2 and TNP 470. Cell counts and 3H-TdR incorporation were measured on culture days 3, 6, and 9.Results On the 9th day, the VECs growth was active in group 1 and accelerated of group 4. The VECs growth was reduced in group 2 ?3 about 1/10 less than that of group 1, and in group 5?6 about 1/7 less than that of group 4 ( P < 0.01 , respectively).Conclusions TNP 470 significantly inhibits the proliferation of VECs of hemangioma cultured with ECGS, or ECGS plus E2. This inhibiting effect is similar to that of hydrocortisone.
