摘要 目的 研究雄激素受体 (AR)亚型在人前列腺移行带与周围带上的分布表达差异及其意义。方法 采用氚标技术与聚丙烯酰胺凝胶等电聚焦电泳的方法 ,对相应组织的AR亚型进行分析。结果 在 14例AR阳性的正常前列腺组织中 ,发现有 4个AR亚型的表达 ,其等电点 (PI)分别在 6.5、6.0、5 .8和 5 .3。在前列腺的移行带和周围带之间有不同的AR亚型表达特征。氚标的双氢睾酮 (DHT)与 4种受体亚型的结合 ,可被非氚标的DHT、睾酮 (T)竞争抑制 ,而孕酮、雌二醇和乙烯雌酚对此却无竞争抑制作用。结论 AR亚型在前列腺带性结构上的不同表达 。 Objective To investigate the differential expression of androgen receptor (AR) isoforms in normal human prostatic peripheral and transitional zones. Methods Using high resolution isoelectric focusing (IEF), the expression of AR isoforms was demonstrated in normal human prostatic peripheral zone and transitional zones.Results Data were obtained from 14 prostatic specimens.Four types of AR isoforms were detected with Pl value at 6.5, 6.0, 5.8 and 5.3.There were different expression rates of AR isoforms in prostatic peripheral and transitional zones. Binding of 3H dihydrotestosterone(DHT) to these four isoforms was inhibited by the addition of 100 fold excess of DHT and testosterone. No effect of progesterone, estradiol and diethylstilbestrol on tritiated hormone binding were observed.Conclusion The expression of AR isoforms is different in prostatic peripheral zone and transitional zone, which may be the molecular basis of various prostatic diseases trending for the occurance in prostatic peripheral zone and transitional zone.
