摘要 目的 探讨粉防己碱 (tetrandrine ,Tet)对肝纤维化大鼠肝组织c fos及c junmRNA表达的影响。方法 皮下注射四氯化碳诱导大鼠肝纤维化模型 ,同时给予不同剂量 ( 5、10、2 0mg/kg)Tet灌胃 ,采用RT PCR法检测肝组织c fos及c junmRNA的表达。结果 不同剂量Tet能不同程度地抑制肝纤维化大鼠肝组织c fos及c junmRNA的表达。 结论 Tet能有效地在转录及其上游水平抑制肝脏胶原合成 ,从而发挥抗肝纤维化作用。 Objective To investigate the effects of tetrandrine (Tet) on the expressions of c fos and c jun mRNA in hepatic fibrotic rats. Methods The hepatic fibrosis model was induced with carbon tetrachloride in rats, and different doses of (5,10 and 20 mg/kg body weight)Tet were given by daily gavage. The levels of c fos and c jun mRNA in liver tissue were determined by RT PCR.Results The levels of c fos and c jun mRNA were significantly higher in the rats of the model group than those of the control group. The increase in levels of c fos and c jun mRNA in drug treated rats(10 and 20 mg/kg body weight) were significantly less than those in rats of the model group. Conclusion Tet can effectively down regulate c fos and c jun mRNA expression, and inhibit collagen type Ⅰ and type Ⅲ synthesis at the transcriptional and its upstream levels.
