摘要
应用甲基纤维素体外半固体培养法观察 32例原发性急性髓性白血病 ( AML )和 4例慢性粒细胞性白血病 -急粒变患者的骨髓或外周血白血病细胞克隆 ( CFU- L)的体外增殖状态 ,以及抗粒 -巨噬系集落刺激因子受体( GM- CSFR) α链单克隆抗体 ( MAb)、抗白细胞介素 - 3受体 ( IL- 3R) α链 MAb、二者共同 β链 MAb对 CFU-L 体外生长的影响。结果显示 :1抗 GM- CSFR、抗 IL- 3R共同 β链 MAb体外可明显抑制 AML 患者 CFU- L 的生长 ( P<0 .0 5 ) ;2抗 GM- CSFRα链 MAb和抗 IL- 3Rα链 MAb对 CFU- L 生长的抑制作用不明显 ( P>0 .0 5 ) ;3各型白血病患者之间对 MAb的反应无明显差异 ( P >0 .0 5 )。认为 GM- CSFR、 IL - 3R的激活在
Using semi solid methylcellulose culture in vitro,the proliferation of colony forming unit leukemia(CFU L) of bone marrow or peripheral blood cells from 32 patients with primary acute myelocytic leukemia (AML) and 4 patients with chronic myelocytic leukemia myeloblastoma,the effects of anti GM CSF receptor α chain,anti IL 3R α chain,anti common β chain monoclonal antibodies (MAb) on growth of CFU L were observed The results showed that①Anti GM CSFR and anti IL 3R common β chain MAb might obviously inhibit the growth of CFU L of patients with AML(P<0 05) ②Anti GM CSFR α chain MAb or anti IL 3R α chain MAb had no effect on growth of CFU L alone (P>0 05) ③There were no statistical difference of the effects of these MAbs on leukemic cell colony growth among AML(P>0 05) These suggest that activation of GM CSFR and IL 3R might play important roles on proliferation of AML blasts
