摘要
目的 研究黄芪成分F3新制剂对人大肠癌CCL -2 2 9,人红白血病K5 62细胞株的抑制作用。方法 实验分为F3单药组、联合用药组、顺铂组 ,另设空白对照组。应用四氮甲唑蓝 (MTT )实验观察细胞的生存、增殖状态 ;应用细胞形态学、基因组DNA电泳、流式细胞仪检测细胞凋亡。结果 一定浓度范围内的黄芪成分F3新制剂直接用药和联合用药均具有明显的生长抑制作用 ,透射电镜下可见凋亡细胞 ;基因组DNA电泳出现“阶梯状”条带 ;流式细胞仪分析出现低于G1期DNA含量的亚二倍体Apoptosis凋亡峰。 结论 黄芪成分F3新制剂对大肠癌CCL -2 2 9及人红白血病K 5
Objective To study the inhibitory effects of F3 new preparation from Astragalus Membranaceus on the cells from carcinoma of the colon(CCL-229)and the K562 line.Methods The viability and multiplication of the cells were observed by methyl thinzoly tetrazolium(MTT) assay.Apoptosis was detected by cell morphology,DNA genome gel electrophoresis and flow cytometry.Results The cytotoxic and inhibitory effects could be apparently observed after administration of F3 new preparation from Astragalus Membranaceus within certain dose range in a manner of either direct administration or combination with others.DNA ladder could be found by DNA genome gel electrophoresis.Apoptosis could be found by electron microscopy.The apoptosis peak of lower quantity of sub-duplex DNA in G 1-phase was observed by flow cytometry.Conclusion F3 new preparation from Astragalus Membranaceus could inhibited the viability and multiplication of the cells from carcinoma of the colon(CCL-229)and the K562 line.
