Survivin在B细胞淋巴瘤中的表达及其与细胞凋亡和预后的关系

Study on expression of survivin gene and the relation with cell apoptosis and prognosis of B esII lymphomas

目的探讨survivin在B细胞淋巴瘤表达及其与细胞凋亡和预后的关系。方法应用TdT介导的dUTP缺口末端标记(TUNEL)技术和免疫组织化学链霉菌抗生物素蛋白-过氧化酶连接法(SP法),检测43例B细胞淋巴瘤中细胞凋亡和survivin蛋白的表达水平。结果survivin在B细胞非霍奇金淋巴瘤表达的阳性率为44.2％(19/43);中度和高度侵袭性淋巴瘤中survivin表达与凋亡指数降低明显相关(P<0.025);弥漫性大B细胞淋巴瘤(DLBCL)中survivin基因的表达高于惰性淋巴瘤(P<0.002);survivin过表达者的平均生存时间明显短于阴性者(P=0.013)。结论 survivin基因的异常表达而引起的凋亡抑制可能在B细胞非霍奇金淋巴瘤,尤其是DLBCL的发生进展中有一定的作用;survivin表达可能是B细胞非霍奇金淋巴瘤的一个新的预后不良因子。

Objective To explore the expression of survivin gene in B cell lymphomas and the relation with cell apoptosis and prognosis Methods Tdt - mediatedUTP nick end labeling and immunochemistry were used to study cell apoptosis and expression of survivin gene in 43 B cell lymphoma samples. Results The survivin positive rate was 44. 2% ( 19/43 ) in B cell of non - Hodgkin. The expression of survivin gene was detected in a significantly greater proportion of intermediately or highly aggressive lymphomas than indolent lymphomas (P <0.025). The expression of survivin inversely correlated with apoptotic index among diffuse large B cell lymphomas (DLBCL) (P<0.002). The mean survival period was significantly shorter in patients expressing survivin than the survivin - negative group. Conclusions Apoptosis inhibition by overex-pression of survivin might be involved in the onset and process of B cell lymphoma, especially of DLBCL. Survivin expression may be a new unfavorable prognostic factor of B cell lymphoma.
