摘要
gp96是存在于真核生物细胞内质网中的分子量约为 96kD的热休克蛋白 (又称GRP94)。它属于HSP90家族 ,是胞质HSP90的旁系同源蛋白。研究证实从小鼠肿瘤组织中分离的gp96注射小鼠后 ,可使小鼠获得针对该肿瘤细胞的特异细胞免疫力。随后发现这种特异性免疫不是由gp96引起 ,而是由其结合的小肽诱发。gp96受体的发现给这种现象的解释提供了线索。人们提出了多种假说来解释这种现象 ,其中一些得到了广泛的支持。另外 ,gp96还参与免疫调节过程。
gp96,a heat shock protein with 96kD molecular weight,reside in the endoplasmic reticulum of eukaryotic organisms.It is the paralog of the cytosol hsp90 and belongs to the hsp90 family.Investigation discovered that when gp96 which derived from mouse maglignant tissue was injected into the same mouse ,it can elicit potent,specific cellular immunity against the tumor.Then it was proved that it is the peptides gp96 chaperoned ,not gp96 that induce specific cellular immunity.The finding of gp96 receptor shed light on this mysterious phenomenon.Scientists all over the world have proposed many possibilities to explain it ,and some of them have received a considerable amount of support .Furthermore,investigator discovered recently that gp96 was also involved in many other immune responses.Completely uncovering the physiological role of gp96 in immunity will be very meaningful for developing new pharmaceutics such as therapeutic vaccines against cancer and virus infection.;
