高效液相色谱法同时测定氯氟滴眼剂中氯霉素和地塞米松磷酸钠的含量

Determination of chloramphenicol and dexamethasone sodium phosphate in eye drops by HPLC

目的 :建立高效液相色谱法同时测定氯氟滴眼剂中氯霉素和地塞米松磷酸钠的含量。方法 :流动相为甲醇 0 .34%磷酸二氢钾 (5 5∶5 0 ) ,流速 1mL·min-1,SpherisorbC18色谱柱 ,检测波长 2 4 0nm。结果 :氯霉素和地塞米松磷酸钠分别在 12 5～10 0 0mg·L-1(r =0 .9992 )、2 5～ 2 0 0mg·L-1(r =0 .9998)内线性关系良好。加样回收率分别为 (10 1.5± 1.93) %、(10 1.0±1.5 9) % (n =3)。日内RSD分别为 0 .6 3% ,0 .5 8% (n =5 ) ,日间RSD分别为 0 .99% ,0 .86 % (n =3)。结论 :该法简单、迅速 ,结果准确可靠。

OBJECTIVE To establish a method for determination of chloramphenicol and dexamethasone sodium phosphate in eye drops by HPLC. METHODS Mobile phase of methanol- 0.34% KH 2PO 4(55∶50) and Spherisorb C 18 were used, the flow rate was 1 mL·min -1 and detection wavelength 254 nm. RESULTS The linear range of chloramphenicol was 125～1000 mg·L -1 (r= 0.9992) and that of dexamethasone sodium phosphate was 25～200 mg·L -1(r= 0.9998). The sample's recoveries were ( 101.5± 1.93)% and ( 101.0± 1.59)% respectively (n=3).The RSD of intra-day were 0.63% and 0.58% (n=5), the RSD of inter-day 0.99% and 0.86% (n=3). CONCLUSIONS This method was proved to be simple,accurate,quick and reliable.
