摘要
目的 :观察TNF受体封闭肽在体外对大鼠腹腔巨噬细胞功能的影响。方法 :用NBT的方法检测呼吸爆发 ;用RT PCR检测大鼠腹腔巨噬细胞IL 1βmRNA和TNF αmRNA水平 ;用Westernblot检测大鼠腹腔巨噬细胞NF κBp6 5核转位。 结果 :TNF受体封闭肽可完全拮抗TNF α诱导大鼠腹腔巨噬细胞呼吸爆发 ,明显抑制TNF α诱导的IL 1βmRNA和TNF αmRNA的转录 ,使腹腔巨噬细胞TNF α诱导的NF κBp6 5核转位明显减少。结论 :TNF受体封闭肽在体外能有效抑制TNF α诱导的大鼠腹腔巨噬细胞功能。
Objective:To observe the effect of TNF receptor(TNFR) blocking peptide in vitro on the functions of rat peritoneal macrophages Methods:Breathe burst was tested by the NBT method; the level of the IL 1β mRNA and TNF α mRNA was determined by RT PCR technique and the nuclear transfer of NF κBp65 was detected by Western blot Results:It was shown that breathe burst of rat peritoneal macrophages induced by TNF α could be completely inhibited by the TNFR blocking peptide The transcription of IL 1β mRNA and TNF α mRNA induced by TNF α was also decreased distinctively by treatment with TNFR blocking peptide which could markedly inhibit TNF α induced nuclear transfer of NF κBp65 Conclusion:TNF receptor blocking peptide could in vitro inhibit effectively TNF α induced activation of rat peritoneal macrophages
