冰冻保存血小板新亚群及其膜表面分子分析 被引量:11

摘要 目的 :探索冰冻血小板新特性的获得与其新亚群膜表面分子变化之间的可能关系。方法 :与新鲜血小板进行比较 ,采用流式细胞术分析冰冻保存血小板膜表面功能分子变化。结果 :根据表达和再表达CD6 2p能力不同可将冰冻血小板分为三个亚群 ,新鲜血小板只有一个明显的亚群。结合磷脂酰丝氨酸 (PS)、CD6 2 p和CD4 2b分子变化可将冰冻血小板分出新的亚群 ,其显著特征包括PS阳性和CD4 2b分子密度显著降低 ;而CD6 2 p阴性亚群对钙离子的激活更为敏感。结论 :血小板通过冰冻保存后 ,产生了新亚群。 Objective:To explore the possible relationshi p of the cryopreserved platelets new characteristics and new platelet subpopulati ons generation Methods: Compared with fresh platelets, Surface functional molecules of cryopreserved platelets were analyzed by flow cytometry Results: The cryopreserved platelets were divided into three subpopulations by CD62p expression and reserve New distinct subpopulations wer e detected by parameters including phosphatidylserine (PS), CD62p and CD42b Th eir distinct characteristic is PS positive and CD42b significant decrease The subpopulation that is both PS and CD62p negative is more sensitive to express CD 62p than fresh platelets when they are stimulated by 2 5 mmol/L Calcium ion Conclusion: After cryopreservation, new subpopulations of platel ets generated Its surface functional molecules lost or altering decided that c ryopreserved platelers in vivo recovery decrease and immediate hemostatic functi on
