摘要
从甘蔗杂种梁河78/121及其亲本华南56/12和崖城58/47共3个材料的幼嫩叶片中提取DNA,使用PCR仪,建立了适合甘蔗PAPD分析的PCR条件,对115个随机引物进行筛选,74个引物可获得甘蔗RAPD多态性,从中找到可用于鉴定该杂种的引物OPR-17,OPK-17和OPR-18。利用该法鉴定甘蔗杂种简单、迅速、可靠,DNA用量少,且不影响被检植株的后期生长。
DNAs were extracted from the young leaves of the sugarcane hybrid 'lianghe 78/121' and its parents 'Huanan 65/12' and 'Yancheng 58/74'. The PCR conditions suitable for RAPD (random amplified polymorphic DNA) analysis of sugarcane were investigated. Of the 115 arbitrary primers used, 74 were polymorphic. The three primers of OPR - 17, OPK - 17 and OPR - 18 were shown to be able to identify hybrids of sugarcane.
