摘要
目的 探讨新生大鼠脊髓神经干细胞 (NSC)移植至血管性痴呆 (VD)大鼠海马后在体内的存活、迁移和分化。 方法 用无血清培养和单克隆技术从新生大鼠脊髓分离具有单细胞克隆能力的细胞群并连续传代培养 ,鉴定为 NSC后 ,将其用 Brd U标记后移植入 VD大鼠海马内 ,利用免疫荧光化学技术检测移植后细胞的生存、迁移和分化。 结果 从脊髓分离的细胞群具有 NSC的特性 ,这些细胞在移植入 VD大鼠海马 8周后能检测到其存在并向四周迁移 ,并且分化成神经元、星形胶质细胞和少突胶质细胞。 结论 从脊髓分离的细胞是脊髓 NSC,将其移植到 VD大鼠海马内能生存。
Objective To study the survival, migration and differentiation of neural stem cells from spinal cord of neogenetic rats after transplanting into hippocampus of adult vascular dementia rats. Methods A cell line which had ability to form clones was isolated from spinal cord of neogenetic rats by using serum\|free cell culture and single cell clone test and passaged continuously. Immunofluorescence was used to detect Nestin antigen of the clone and specific antigen of mature neural cell after the clone differentiated. The cell line was labeled before transplanted into hippocampus of adult vascular dementia rats. Immunofluorescence was used to detect survival, migration and differentiation of BrdU\|labeled cells. Results The cell line isolated from spinal cord of neogenetic rats expresses Nestin antigen and has the potential to form clones and differentiate into neurons, astrocytes, and oligodendrocytes. These cells which were transplanted into hippocampus of vascular dementia rats eight weeks before can be detected, which indicates they can survive, migrate and differentiate. Conclusion The cell line which expresses Nestin antigen is multipotent cell line and has ability to self\|renew, and believed that it contains stem cells of spinal cord. When transplanting them into the hippocampus of vascular dementia rats, they can survive, migrate and differentiate.
