摘要
目的 探讨胃主要连接蛋白基因 (connexin 32 )在不典型增生和胃癌的表达及其与胃癌发生发展的关系。 方法 运用原位杂交方法和免疫组织化学方法检测 Cx32 m RNA及蛋白和增殖细胞核抗原 (PCNA)在 70例原发性胃癌、6 2例不典型增生和 16例正常胃粘膜中表达。 结果 在正常胃粘膜、不典型增生和胃癌中 ,Cx32和 Cx32m RNA的表达水平逐步下降 ;PCNA标记指数分别为 0 .75 %± 1.5 2 % ,13.0 6 %± 16 .0 9%和 38.0 9%± 2 1.89%。三者在不典型增生、胃癌中表达与正常组相比差异有显著性 ,不典型增生组与胃癌组比较差异也具有显著性 (P <0 .0 5 )。Cx32 m RNA与 Cx32表达具有一致性 (P<0 .0 1)。Cx32表达与 PCNA L I呈负相关 (P<0 .0 1)。 结论 Cx32基因在不典型增生和胃癌中表达逐步下降 。
Objective To investigate the relationship between carcinogenesis of stomach and the expression of connexin 32(Cx32) and its significance. Methods The expression of Cx32 and proliferative cell nuclear antigen(PCNA) were detected by immunohistochemistry, and Cx32 mRNA by in situ hybridization in 70 cases of gastric cancer, 62 dysplasia and 16 normal gastric mucosa. Results In normal gastric mucosa, dysplasia and neoplasms, the positive rates of Cx32 were 100%, 90 3% and 18 6%, Cx32 mRNA were 93 7%, 77 4% and 12 9% and PCNA label index(LI) were 0 75%±1 52%, 13 06%±16 09% and 38 09%±21 89%, respectively. Significant differences of both Cx32 mRNA and Cx32 expression existed among gastric cancer, dysplasia and normal mucosa(P<0 05). The expression of Cx32 is inversely correlated with PCNA LI(P<0 01). Conclusion Gastric cancer and dysplasia exhibited lower positive rates of Cx32 mRNA and Cx32 compared with that of normal gastric mucosa. It is suggested that lower expression of Cx32 play an important role in the development of gastric carcinoma.
