环氧化酶-2、bcl-2与bax在胃癌的表达及意义

Expression and Significance of cyclooxygenase-2, bcl-2 and bax in Human Gastric Carcinoma

目的 研究环氧化酶 - 2 (cox- 2 )与 bcl- 2、bax在胃癌中的表达及其与胃癌细胞凋亡的关系。 方法 采用SP免疫组织化学法检测胃癌及癌旁组织各 6 0例标本中 cox- 2、bcl- 2、bax的表达 ,用脱氧核糖核酸末端转移酶介导的d UTP缺口末端标记法 (TUNEL )检测胃癌及癌旁组织的凋亡率。 结果 胃癌组织的 cox- 2、bcl- 2的表达明显高于癌旁组织 (P<0 .0 5 ) ,bax的表达明显低于癌旁组织 (P<0 .0 5 ) ;cox- 2阳性组胃癌组织的凋亡率显著低于阴性组 ;cox-2的表达与 bcl- 2呈正相关 ,与 bax呈负相关。 结论 cox- 2、bcl- 2、bax的异常表达与胃癌细胞凋亡抑制有关 ;cox- 2能够上调 bcl- 2的表达并下调

Objective To study the expression of cyclooxygenase\|2(cox\|2), bcl\|2 and bax gene in human gastric carcinoma tissues and the relationship between cox\|2, bcl\|2, bax and the apoptosis of gastric carcinoma cells. Methods S\|P immunohistochemistry was used to detect the expression of cox\|2, bcl\|2 and bax in 60 cases of gastric carcinoma and their adjacent noncancerous tissues, and terminal deoxynucleotidyl transferase\|mediated dUTP nick end labeling was used to detect the apoptosis in above tissues. Results The positive rates of cox\|2 and bcl\|2 in gastric carcinomas were significantly higher than those in the adjacent noncancerous tissues(P<0 05). The positive rate of bax in gastric carcinomas was significantly lower than the adjacent noncancerous tissues(P<0 05). The index of apoptosis in gastric carcinomas was significantly lower than the adjacent noncancerous tissues(P<0 05). Conclusion The apoptosis of gastric carcinoma cells correlated with the expression of cox\|2, bcl\|2 and bax. The high expression of cox\|2 up\|regulation the expression of bcl\|2 gene and down\|regulation of bax expression made the inhibition of apoptosis of gastric carcinoma cells.
