摘要
根据猪的消化生理特点和饲喂饲料,确定猪用复合酶以酸性蛋白酶、木聚糖酶为主,兼有纤维素酶,并采用单因素搜索试验对培养基组成、培养条件进行优化.实验确定的最佳培养基组成为:麸皮60 g,花生壳粉40 g,豆粕10 g,(NH4)2SO4 2 g,MgSO4 0.05 g,K2HPO4 0.1 g.最佳培养条件为:接种量w(曲种)=0.3%,每g原料含3×105个孢子,培养温度30℃~35℃和适宜的培养时间.在此条件下,所产酶活力分别为:纤维素酶8597 u/g,木聚糖酶5054 u/g,酸性蛋白酶861 u/g,糖化酶2110 u/g.经在猪饲料中应用效果良好.
According to digestion physilogical characteristic and feeds of pigs, acid proteases, xylanases and cel-lulases were confirmed as composition of compound enzymes employed to pig feed. Compositions of medium and cultivating conditions were optimizated by single factor test. The optimum medium compositions: wheat bran 60 g,peanut causing 40 g.cake bean 10 g, (NH4)2SO42 g,MgSO4 0.05 g,K2HPO4 0.1 g.The optimum cultivating conditions:the volume of inoculum is 0.3% and 3 × 10 cells/g material, the temperature is 30 ℃- 35℃ and selects optimal time.In this way,it could produce enzymes asicellulase activity 8597 u/g,xylanase activity 5054 u/g,acid protease activity 861 u/g,gluvoamylase activity 2110 u/g.It is better on application of pig's food.
