番茄外植体诱导直接分化不定芽建立高频再生系统 被引量:35

摘要 研究以番茄(LycoperciconesculentumMill)栽培品种贵妃和碧娇为试材,通过对番茄子叶、下胚轴、叶片和茎段进行离体培养,不经愈伤组织阶段,诱导直接分化不定芽,研究并得到其高频再生的最适培养基和试管苗扦插繁殖的最适培养基,并对影响高频再生的其他因素做了研究,建立了再生频率高达98%以上的番茄高频再生系统,为番茄快速繁殖及基因转化研究奠定了基础. Tomato(Lycopercicon esculentum Mill) Cultivars,Guifei and Bijiao,were used as the materials in the study. The cotyledon, hypocotyl, leaf and stem of tomato were cultured in vitro and induced directly to differentiate buds.The optimal medium of highfrequency regeneration was investigated and obtained.The optimal medium of cutting breeding of tomato testtube seedlings was also obtained, the other factors affecting the highfrequency regeneration were also studied, and the highfrequency regeneration system of tomato was established with the regeneration frequency being above 98%, which became the important base of the highfrequency transformation of tomato.
