高效低成本生产健壮马铃薯试管苗技术研究

摘要 为了简化马铃薯脱毒试管苗培养方法,降低生产成本,进行了调整培养基成分、试管苗复壮和改进实验设备三方面研究.结果表明,以食用棉白糖、倍力凝作为蔗糖、琼脂的替代物,再将微量元素减半,用经过滤的自来水配制的培养基对马铃薯苗的生长不会产生负面效应,培养成本可降低84.4%.对生长势弱的马铃薯苗,附加2mg/L IBA的2倍MS液体培养基对其有促壮作用.用回收旧罐头瓶作培养容器,用聚丙烯膜、线绳封口,不仅对试管苗的生长无任何影响,而且可节约投资达90%以上. In order to simplify culture methods,to reduce the production cost,to improve the prac- ticability of technique,three aspects,adjusting the components of medium and dosage and culturing vig- orous potato plantlets and selecting low—cost equipment were studied.In the medium,half MS micro— composition was used,the sucrose and agar and ditilled water were substituted by sugar and polygel and filtered tap water.The results indicated that potato plantlets grew well in the modified medium,,and the medium cost could be reduced by 84.4%.Adding 2mg/L IBA to double MS liquid medium,in which the leggy and feeble potato plantlets could become vigorously,modifying equipments such as recy- cling glass can battles as culture containers,using polyacrylonitrile film and cotton threads to seal,the cost could be reduced by 90%,and this culture method didn't cause negative impact on potato plantlets.
