摘要
目的观察应用直流电疗法对肺癌细胞凋亡的影响,研究直流小电场对于肺癌细胞凋亡的诱导作用及其可能机制。方法应用体外直流小电场作用于肺腺癌细胞系A549,观察其对肺癌细胞凋亡和重要基因p53、Rb以及E2F1表达的影响。结果直流小电场对肺癌细胞系A549有明显的诱导作用,200mV/mm和250mV/mm组癌细胞大量脱落,电场组凋亡癌细胞数目随电场强度升高而增多(F=795.372,P=0.000)。电场组p53、Rb基因mRNA表达明显升高,E2F1基因mRNA表达明显下降。结论直流小电场对肺癌细胞系A549具有明显的凋亡诱导作用并可能与p53、Rb以及E2F1基因的表达相关。
Aim To observe the effect of direct current (DC) therapy on apoptosis of lung cancer cells and study the inducement and mechanism of small DC electric field in the apoptosis of lung cancer cells.Methods Small DC electric field stimulation (EFs) were applied in human lung cancer cell line A549 in vitro to observe the effects on apoptosis of cancer cells and the mRNA expressions of p53, Rb and E2F1 genes by reverse transcriptase polymerase chain reaction (RT PCR).Results A great amount of cancer cells were shed from culture dish in 200 mV/mm and 250 mV/mm groups and the number of the apoptotic cancer cells significantly increased along with the improvement of intensity of electric field.(F=795.372,P= 0.000). The mRNA expression level for p53 and Rb genes increased while that for E2F1 gene decreased significantly.Conclusion The small direct current EF induces the apoptosis of lung cancer cells significantly, probably correlated with the expressions of p53, Rb and E2F1 genes.
