摘要
用SDS-聚丙烯酰腔凝胶电泳(SDS—PAGE)法,观察了家蚕不同发育阶段血液主要血浆蛋白质浓度的发育变化,比较了不同的地方性三眠蚕品种间主要血浆蛋白质组成及浓度的异同。比较家蚕与野桑蚕、蓖麻蚕、樗蚕及柞蚕血浆的电泳图谱,发现家蚕与野桑蚕的电泳图谱相似而不同于蓖麻蚕、樗蚕和柞蚕。后三者之间的电泳图谱很相似,它们不存在类似于家蚕及野桑蚕中的分子量为30KD的一组蛋白质,但也存在分子量相当于家蚕贮藏蛋白质的成分。用精制的家蚕贮藏蛋白质、30K蛋白质和卵黄磷蛋白分别制备免抗血清,并使之与五种试验蚕的血液分别进行了双向免疫扩散试验。
Authors observed developmental changes in the hemolymph concentratioms of major plasma proteins of differnt developmental stages in the Bombyx mori by SDS-polyacrylamide gel electrophoresis. Moreover, the differences of components and concentrations of major plasma protiens in the local three moulter varieties were compared.
By comparing the plasma electrophoresis maps, it was found that the map of Bombyx mori is similar to the map of Bombyx mandarina and is different from the maps of Philosamia cynthia ricini, Philosamia cynthia and Antheraea pernyi.The electrophoresis maps of the last 3 species of silkworms are similar reciprocally, there isn't a group of proteins whose molecular weight is 30KD in the Bombyx mori and Bombyx mandarina but there is a component whose molecular weight is equal to the Bombyx mori storage proteins in the last 3 species of silkworms.
Rabbit antibodies to the purified storage proteins,30KD proteins and Vite-llin were prepared and double immunodiffusion has been carried out with the hemolymph from 5 species of silkworms.
