高压氧对脑缺血再灌注小鼠血小板膜糖蛋白CD31、CD61和CD62p的影响 被引量:4

Effects of hyperbaric oxygen on the expression of platelet membrane glycoprotein-CD31,CD61 and CD62p in mice after cerebral ischemia-reperfusion

摘要 目的 研究高压氧 (HBO)对脑缺血再灌注小鼠血小板膜糖蛋白的影响。方法 小鼠随机分为 HBO组、高气压组、单纯缺血再灌注组、假手术组和正常对照组。夹闭小鼠双侧颈总动脉 ,建立脑缺血再灌注动物模型。应用流式细胞术分别检测再灌注后第 1,3,5天的血小板膜糖蛋白 CD31、CD6 1和CD6 2 p的表达。结果  HBO组第 1,3天的血小板膜糖蛋白 CD31表达水平显著低于同期再灌注组 (均 P<0 .0 5 ) ;HBO组第 1天的 CD6 1、CD6 2 p表达水平显著低于同期再灌注组 (均 P<0 .0 5 )。 HBO组、高气压组、单纯缺血再灌注组第 5天的血小板膜糖蛋白 CD31、CD6 1和 CD6 2 p表达均恢复正常水平。结论 早期 HBO治疗可有效抑制缺血再灌注后血小板膜糖蛋白 CD31、CD6 1和 CD6 2 p的表达 ,可能有助于减轻再灌注损伤。 Objective To study the effects of hyperbaric oxygen on platelet membrane glycoprotein in mice after cerebral ischemia-reperfusion.Methods The model mice were randomly divided into hyperbaric oxygen group,hyperbaric air group, simple ischemia-reperfusion group,sham operation group and control group.Cerebral ischemia-reperfusion models were established by clamping both common carotid arteries in mice.Using flow cytometry,the expression of CD31,CD61 and CD62p on the platelet membrane in the model mice were measured,respectively on the 1 st,3 rd,5 th day after reperfusion.Results The expression of CD31 on platelet membrane in the hyperbaric oxygen group on the 1 st and 3 rd day was significantly decreased,compared with the simultaneous simple ischemia-reperfusion group(respectively P<0.05,P<0.05).The expression of CD61 and CD62p in the hyperbaric oxygen group on the 1 st day was significantly decreased,compared with the simultaneous simple ischemia-reperfusion group (respectively P<0.05,P<0.05).The expression of CD31,CD61 and CD62p in the hyperbaric oxygen group,the hyperbaric air group and the simple ischemia-reperfusion group on the 5 th day all returned to the common level on the 5 th day.Conclusions Early hyperbaric oxygen therapy can inhibit effectively the expression of CD31,CD61 and CD62p on platelet membrane in ischemia-reperfusion mice,which may relieve the reperfusion injury.
