摘要 目的 :观察急性肺损伤 (ALI)小鼠T辅助淋巴细胞 (TH)TH1/TH2的平衡变化 ,探讨TH1/TH2平衡变化在ALI发生中的意义。方法 :2 4只BLAB/C小鼠随机分成ALI组、对照组。酶联免疫吸附法 (ELISA)测定脾细胞培养上清液中干扰素 (IFN) γ、白细胞介素 (IL) 4蛋白含量 ,半定量逆转录聚合酶联反应 (SqRT PCR)方法进一步观察脾细胞中IFN γmRNA、IL 4mRNA表达。伊文思蓝荧光法反映肺血管通透性改变。肺湿重 /干重比反映肺水量。HE染色观察肺病理变化。结果 :与对照组比较 ,ALI组脾细胞中IFN γ、IFN γmRNA表达均显著下降 (P <0 .0 1)。IL 4、IL 4mRNA明显升高 (P <0 .0 1)。ALI组肺组织中伊文思蓝浸出量、肺湿重 /干重比均明显升高。结论 :ALI时 ,TH1向TH2漂移 ,提示炎症与抗炎症反应失衡 ,抗炎症反应增强。 Objective: To observe the variation of the balance of TH1/TH2 cytokine on the condition of acute lung injury. Methods: Mice were divided randomly into control saline group and acute lung injury group( n =12 per group). By using enzyme linked immuno sorbent assay (ELISA), Interferon(IFN) γ?interleukin(IL) 4 in supernatants of splenocytes were quantified, meanwhile cytokine mRNA expression was determined by Semi quantitatively reverse transcriptase polymerase chain reaction(SqRT PCR). The pulmonary extravascular lung water were determined by lung wet to dry weight ratio. The quantity of protein immersed were measured by Evan's blue fluorescence, the character of lung pathology was observed by using HE dying. Results: As compared with control group, splenocyte cytokine IFN γ and IFN γmRNA expression were decreased( P <0.01), IL 4 and L 4mRNA expression were increased( P <0.01) in ALI group. The lung wet to dry weight ratio and quantity of lung Evan's blue immersed were increased markedly in ALI group. Conclusion: A shift to a TH2 type splenocyte cytokine response occurs after ALI, illustrating inflammatory and anti inflammatory response lose balance on the condition of ALI with the anti inflammatory response enhaneed.
