大麦再生植株遗传稳定性检测方法的建立和应用 被引量:1

摘要 为了解大麦离体培养中影响遗传稳定性的因素,对大麦3个品系的花粉再生植株的诱导培养采用了不同温度、不同激素浓度及不同低温预处理的培养方式,对各种试验条件下诱导的再生植株进行了过氧化物酶同工酶和酯酶同工酶酶谱的分析。结果表明,在培养温度为30℃的酶谱条带较常规培养温度25℃的酶谱条带有较大变化;在培养基激素2,4-D为5mg/L浓度的酶谱条带较常规激素浓度2.5mg/L的酶谱条带也有较大变化;而低温预处理也不同程度地导致了酶谱的变化。根据同工酶酶谱分析,认为在大麦再生植株的诱导培养过程中,采用常规的25℃培养温度和培养基激素2,4-D为2.5mg/L浓度的处理可以保持材料本身的遗传稳定性;而提高培养温度、培养基激素浓度及采用低温预处理的措施有可能影响其遗传稳定性的表达。 To know effecting factor of culture in virto barely' s hereditary stability,three barely lines' s pollen regeneration plant were cultured in different temperature,different endocrine consistencies and different low-temperature pretreatments.The peroxi-dase and esterse iszoymes of three regeneration plants were analysed.The results showed that the bands of emzyme pattern at 30C culture temperature changed more than that at 25C culture temperature which was the regular culture temperature,and the bands of 5mg/L endocrine 2,4-D changed more than that of 2.5mg/L endocrine 2,4-D which was the regular concenteration,and the changes of esterase pattern were influnced by the low-temperature pretreatments.According to analysis of isozymes pattern,we had a view of that if we rose the culture temperature and increased the endocrine 2,4-D's consiatency,and dealed with low-temperature pretreatment,hereditary stability of barely regeneration plants would be effected.So we suggest that 25^ culture temperature and 2.5mg/L endocrine 2,4-D should be fit to culture.
