摘要
目的 :观察黄芪对肝星状细胞有关胶原降解基因表达的影响。方法:选择肝星状细胞系作为体外研究对象 ,采用MTT法选择药物的实验浓度 ,酶联免疫测定 (ELISA)法检测细胞上清液中Ⅰ、Ⅲ、Ⅳ型胶原含量 ,逆转录多聚酶链反应检测间质性胶原酶及金属蛋白酶组织抑制因子基因表达水平。结果:与对照组比较 ,黄芪组细胞上清液中Ⅰ、Ⅲ、Ⅳ型胶原含量明显降低(P<0.01),黄芪组间质性胶原酶基因表达水平明显增强 (P<0.01) ,而金属蛋白酶组织抑制因子基因表达水平无明显差异 (P>0.05)。
Objective: To study the effects of Astragalus on gene expression for collagen degradation of hepatic stellate cell in vitro.Methods: Hepatic stellate cell line(HSC-T6) was cultured in vitro. Concentration of Astragalus was selected by MTT colorimetric assay . Collagen type I,Ⅲ,Ⅳ contents in cell media were detected by ELISA. The gene expression levels for interstitial collagenase and tissue inhibitor of metalloproteinase-1(TIMP-1) were measured by reverse transcription polymerase chain reaction.Results: Compared with control group, collagen type I,Ⅲ,Ⅳ contents in cell media of herb group were significantly decreased (P<0.01),gene expression level of interstitial collagenase in Astragalus group was significantly enhanced(P<0.01).On the other hand, no significant difference between two groups in gene expression level of TIMP1(P>0.05).Conclusion: Astragalus can prevent liver fibrosis via enhancing gene expression of interstitial collagenase.
