摘要
目的 :探讨TH01、TPOX基因座在天津汉族群体的遗传多态性和法医学应用价值。方法:EDTA抗凝血样采自天津地区无血缘关系个体 ,TKM法提取DNA,PCR扩增、聚丙烯酰胺垂直板电泳分离及银染技术 ,对天津汉族人群164例无关个体TH01、TPOX基因座分型。结果:TH01检出5个等位基因 ,14种基因型 ;TPOX共检出5个等位基因 ,14种基因型。两基因座基因频率分布符合Hardy -Weinberg平衡。TH01、TPOX基因座观察杂合度分别为0.6585、0.6281,个人识别机率分别为0.8174、0.8137。结论:TH01。
Objective: To understand the distribution of genes and genotypes of TH01 ?TPOX loci in Han population in Tianjin.Methods:EDTA-blood specimens were collected from the unrelated individuals. The DNA samples were extracted with TKM method and were amplified by PCR technique. Polyacrylamide gel electrophoresis followed by silver staining were used to analyze the polymorphism of TH01 TPOX loci.Results:5 alleles and 14 genotypes of TH01, 5 alleles and 14 genotypes of TPOX were observed. The genotype distributions in two loci were in accordance with Hardy-Weinberg equilibrium.The heterozygosities observed (H)were 0.658 5?0.628 1 for TH01 and TPOX respectively.The discriminating powers(DP) were 0.817 4?0.813 7. Conclusion: The results demonstrated that TH01 and TPOX locus is highly polymorphic and can be used inforensic identification and genetic research of population as well.
