临床乳腺癌细胞周期蛋白非时相性表达的初步探讨

摘要 目的 初步探讨临床乳腺癌细胞的细胞周期蛋白 (Cyclin)表达规律并予以归类 ,为进一步研究肿瘤的细胞周期机制提供依据。方法 采用Cyclin/DNA双参数流式细胞术对临床上获取的乳腺癌细胞的Cyclin (CyclinD1、E、A和B1)和DNA表达进行检测。结果 在 16例乳腺癌标本中 ,细胞周期蛋白的非时相性表达可以归类如下 :①Ⅰ型CyclinD1、E、A和B1均呈非时相性过表达 (5例 )。②Ⅱ型CyclinsD1、E、A和B1均呈低表达或不表达 (3例 )。③Ⅲ型CyclinD1呈过表达并贯穿于整个细胞周期 ,而CyclinsE、A和B1呈低表达或不表达 (3例 )。④Ⅳ型CyclinsE、A和B1呈不同程度的非时相性过表达 ,而CyclinD1呈低表达或不表达 (5例 )。结论 Cyclins在乳腺癌细胞中的非时相性表达存在着一定的规律 。 Objective To systematically analyze the characteristic and rule of unscheduled Cyclins expression in breast cancer cells (Cyclins D1, E, A, B1), so as to provide a reliable foundation for further researching the mechanism of cell cycle broken in breast cancer. Methods The samples from clinical breast cancer were harvested and the expression of Cyclins (D1, E, A, B1) was detected by using Cyclins/DNA bivariate flow cytometry. Results There were 4 types of unscheduled Cyclins expression in all 16 samples from clinical breast cancer: ① TypeⅠ, unscheduled and high expression of Cyclins D1, E, A and B1 (n=5); ② TypeⅡ, low (or no ) expression of Cyclins D1, E, A and B1 (n=3); ③ Type Ⅲ, Only Cyclin D1 over-expression throughout the cell cycle, but much low expression of Cyclins E, A and B1 (n=3); ④ Type Ⅳ, different unscheduled over-expression of Cyclins E, A and B1, but low expression of Cyclin D1 (n=5).Conclusion Each type of unscheduled Cyclin expression in breast cancer cells has its own mechanism of cell cycle broken in tumorigenesis.
