槲皮素逆转柔红霉素在耐药细胞株K562/ADR内的异常分布

摘要 目的 研究槲皮素处理前后柔红霉素 (DNR)在耐药细胞株K5 62 /ADR亚细胞结构的不同分布。方法 利用蒽环类抗生素固有的荧光 ,通过共聚焦激光扫描显微镜观察槲皮素作用后DNR在耐药细胞株K5 62 /ADR内亚细胞结构分布的改变。结果 与DNR在敏感细胞株K5 62 /S细胞核、胞浆均匀分布不同 ,在K5 62 /ADR细胞 ,DNR主要集中在核周及周边胞浆 ,核内DNR荧光很少 ;经槲皮素处理后 ,可恢复DNR在K5 62 /ADR内细胞核、胞浆中的弥漫分布。结论 DNR在耐药细胞中的异常分布与肿瘤细胞耐药的形成有关 。 Objective To investigate the restoration of the subcellular distribution of daunorubicin in the P glycoprotein mediated multidrug resistant cell line K562/ADR by quercetin. Methods The effect of quercetin on the subcelluar distribution of DNR in K562/ADR was under taken by confocal scanning laser microscopy (CSLM). Results Compared with the drug sensitive cell K562/S in which DNR fluorescence appeared diffusely in the nucleus and cytoplasm, DNR in K562/ADR cells was distributed at the perinuclear region and peripheral cytoplasm. Quercetin could markedly restore the diffuse cytoplasmic and nuclear fluorescence distribution in the resistant cell line. Conclusion Quercetin can restore the altered subcellular disposition of DNR in resistant cell line, which is involved in the mechanism of multidrug resistance.
