局部肾素血管紧张素系统在人T淋巴细胞增殖中的表达及作用

摘要 目的 研究局部肾素血管紧张素系统 (RAS)在人T淋巴细胞增殖中的表达情况及其与T淋巴细胞增殖的相互关系。方法 分离、纯化、激活和培养人外周血T淋巴细胞。采用放射免疫法测定人外周血T淋巴细胞培养上清液中血管紧张素转化酶 (ACE)、AT1受体 (AT1R)、肾素和血管紧张素 (ANG)Ⅱ的水平 ,及其在T细胞增殖过程中的表达变化 ;采用RT PCR半定量测定ACE、AT1R和AT2RmRNA ;采用免疫组织化学法测定AT1R表达 ;采用3H 胸腺嘧啶摄入法测定外源性ANGⅡ及其特异阻断剂对T细胞增殖的直接作用。结果 T淋巴细胞表达RAS各种组分 ,在T淋巴细胞增殖过程中存在RAS上调现象 ,RAS组分mRNA表达增加 ,细胞上清液中RAS组分水平增加。ANGⅡ对T淋巴细胞增殖有直接促进作用。结论 T淋巴细胞存在RAS的各种组分 。 Objective To study the expression of regional renin angiotensin system(RAS) in the human T lymphocyte and its regulatory role in T lymphocyte proliferation. Methods After separation and purification, the human peripheral T lymphocytes were activated by PHA P and cultured in medium containing IL 2. AT1 receptor expression was detected by immunohistochemistry. RAS component (ACE, AT1R and AT2R), mRNA levels were measured by semi quantitative RT PCR, ACE, renin and angiotensinⅡ(ANG Ⅱ) levels in culture supernatant in term by radioimmunoassay respectively. Exogenous ANG Ⅱ and its antagonist were used to study the direct regulatory effect on T lymphocyte proliferation. Results T cell contained RAS component, and showed upregulation of RAS during T cell proliferation: RAS mRNA expression and RAS component in culture supernatant were increased. Exogenous RAS have direct stimulatory effect on T lymphocyte proliferation. Conclusion T lymphocyte contain components of RAS, and RAS is closely related to T lymphocyte proliferation.
