D-聚甘酯片延长人体aPTT时间药效动力学研究 被引量:6

摘要 目的:研究D-聚甘酯片在人体内延长部分凝血活酶时间(aPTT)的时效和量效关系。方法:通过9名健康成年男性志愿者分别依次单次口服400、500、600mg的D-聚甘酯片后,应用免疫荧光比浊法测定给药后不同时间抽血测定aPTT值,计算药效动力学参数。结果:D-聚甘酯能显著延长人体aPTT,并于给药后1h达高峰,效应持久,并随剂量的增加,aPTT最大延长效应明显,呈剂量依赖性。结论:D-聚甘酯延长人体aPTT有显著的时效和量效关系。 Objective To study the relationship of time-effect and dose-effect of active part thromboplastin time (aPTT) elongation caused by D-polymannuronicate in vivo. Methods The aPTT of plasma sample collected from the healthy male volunteers after taking orally 400、500、600mg D-polymannuronicate respectively by turns was determined by the immuno-fluorescence turbidity method, and the pharmacodynamics parameters were calculated. Results aPTT could be extended significantly by D-polymannuronicate in vivo , and reached the peak at 1h after administration. The effect of aPTT elongation was lasted for a long time, and was significantly dose-dependent with the increase of the dose. Conclusion The relationship of time-effect and dose-effect of active part thromboplastin time(aPTT) elongation caused by D-polymannuronicate in vivo was considered to be significant.
