摘要
用 8 0 %乙醇溶液浸提饲料中的越霉素A ,浓缩后分别用薄层色谱法对 6种展开剂、3种薄层板类型和 3种显色剂进行了筛选。研究结果表明 ,用 80 %乙醇溶液提取越霉素A、以氯仿∶甲醇∶氨水∶水 =1∶4∶2∶2为展开剂、在碱性硅胶G薄层板上展开、用 0 .2 5 %茚三酮乙醇溶液显色 ,能将越霉素A与其他活性成分在薄层板上分离 ,越霉素A的Rf值为 0 .4 7。该法的最低检测浓度为 0 .0 4mg/ml,最低检出限为 0 .2 μg。该方法简单 ,易操作。
A selective and accurate method of determination of destomycin A(DM. A) in feeds is described. DM. A in feed was extracted with 80% ethyl alcohol and was separated with other components in feed by thin layer chromatogrophy on alkalinity silica gel G plates,and was developed in the mobile phase with trichloromethane∶ methanol∶ammonium hydroxide ∶ water=1 ∶4 ∶ 2 ∶ 2, then was displayed with 0.25% ninhydrin . The Rf of DM. A in feed is 0.47. The separation and detection procedure are easy to perform.
