摘要
目的 建立新疆 3个不同地理群体的中麻黄 (Ephderaintermedia)指纹图谱 ,进行遗传多样性分析。 方法 通过 2 0个1 0碱基随机引物对新疆 3个不同地理群体的中麻黄进行PCR扩增 ,并用人工栽培种作为对照 ,通过计算遗传相似性系数 ,建立UPGMA聚类图。结果 共扩增出 1 53个多态位点 ,建立了它们的基因组DNA指纹图谱。结论 不同地理群体中存在丰富的遗传多样性 ;相距越远 ,群体间相似性程度越低 ;
OBJECTIVE: To establish the chromosome DNA fingerprints of Ephdera intermedia from three local populations in Sinkiang and to analyze their genetic diversity. METHODS: Twenty different 10 bp oligonucleotide primers of arbitrary sequence were used to amplify with the cultivated specie as control. By calculating the genetic similarity coefficient, UPGMA clustering map was set up. RESULTS: A total of 153 polymorphic bands were amplified and the chromosome DNA fingerprints of these local populations were established. CONCLUSION: There was rich genetic diversity in different local populations. The far the genetic distances, the lower the comparability degree among populations. And the similar genetic characteristic lies in both the wild specie and the cultivated specie in the same location.
