晶状体上皮细胞凋亡及bcl-2、bax表达与年龄相关性白内障形成的关系 被引量:12

摘要 目的 :探讨晶状体上皮细胞凋亡与年龄相关性白内障发生的关系。方法 :用透射电镜观察 2例年龄相关性白内障患者和 1例正常人晶状体上皮细胞的超微结构。用脱氧核苷酸末端转移酶缺口标记原位细胞检测法 (theterminaldeoxynucleotidyltransferasa(TdT) medinteddUTPnick endlabeling,TUNEL)检测晶状体上皮的凋亡细胞。采用链霉菌亲生物素蛋白 过氧化物酶标免疫组织化学法 (streptavidin peroxi dase,SP)染色 ,镜下检测 4 6例年龄相关性白内障患者和 13例健康成年人、7例胚胎的晶状体上皮细胞的bax基因与bcl 2基因的表达情况。结果 :在年龄相关性白内障患者的晶状体上皮细胞的超微结构中可见凋亡之细胞 ,健康成年人晶状体上皮细胞正常。年龄相关性白内障患者的晶状体上皮细胞凋亡百分率为 2 1.2 0 % ,健康成年人的晶状体上皮细胞凋亡百分率为 0 .2 5 %。年龄相关性白内障患者晶状体上皮细胞的bax基因表达为 10 0 % ,bcl 2基因表达为 0 % ,健康成年组和胚胎晶状体上皮细胞的bax基因表达为 0 % ,bcl 2基因表达为 10 0 %。结论 :晶状体上皮细胞的凋亡在年龄相关性白内障的发生中起重要作用。 Objective:Age related cataract is a major ocular disease that causes blindness in many countries of the world. In spite of more than fifty years of basic and clinical research,there is no nonsurgical intervention to prevent or treat age related cataract,but there is a better understanding of the manifold complexities of this age related condition. To evaluate the relationship between damage from lens epithelial cell death and apoptosis in patients with age related cataract.Methods:Transmission electron microscopy was used to observe the apoptotic morphology of lens epithelial cells in 2 cases with an age related cataract and 1 normal eye. Terminal deoxynucleotidyl transferasa(TDT) mediated dUTP nick end labeling(TUNEL) assays were done using anteriorless capsules obtained in surgery from 46 patients with age related cataract and from 13 normal patients and 7 embryos. The expression of bcl 2 and bax was detected by the immunohistochemical streptavidin peroxidase(SP) method.Results:The ultrastructure of the lens epithelial cell showed that apoptotic cells were found in age related cataract,while few were found in the control lens epithelial cell. The TUNEL method revealed that the percent of apoptosis in the lens epithelial cell was 21.20% in age related cataract,while the percent of lens epithelial cell apoptosis was 0.25% in normal eyes. The percent of bax positive cells was 100% in age related cataract,while the percent of bax positive cells was 0% in normal eyes and in embryos. The percent of bcl 2 positive cells was 0% in age related cataract,while the percent of bcl 2 positive cells was 100% in normal eyes and in embryos.Conclusion:These results suggest that apoptosis of lens epithelial cells may play an important role in the development of age related cataract.
