重组人α干扰素对瘢痕成纤维细胞增殖的生物学效应 被引量:1

BIOLOGICAL EFFECTS OF RECOMBINANT HUMAN INTERFERON α ON THE PROLIFERATION OF HUMAN SCAR FIBROBLASTS

摘要 目的 观察重组人α 干扰素 (rHuIFN α)对培养的人瘢痕成纤维细胞增殖的影响 ,探讨rHuIFN α对瘢痕成纤维细胞生物学作用。方法 将rHuIFN α加入瘢痕成纤维细胞培养基内 ,采用MTT法测定细胞增殖率 ,用3H TdR掺入实验检测细胞DNA合成率 ,应用流式细胞仪分析细胞增殖周期。结果 当rHuIFN α浓度 >40 0 0U/ml时 ,实验组OD值明显低于对照组 (P <0 0 1)。rHuIFN α组3H TdR掺入率明显下降 ,与DMEM组比差异显著 (P <0 0 1)。rHuIFN α组培养 2 4h时瘢痕成纤维细胞进入S期的比例明显降低 (P <0 0 1) ,至 72h时已无显著差别。结论 rHuIFN α可以抑制瘢痕成纤维细胞的DNA合成及增殖 ,其机理为rHuIFN α对瘢痕成纤维细胞进入有丝分裂S期的抑制 。 Objective: To observe the biological effects of recombinant human interferon α (rHu IFN?α) on hypertrophic scar fibroblasts. Methods: rHu IFN?α was added to the cultivated hypertrophic scar fibroblasts. MTT assay was used to test the effects of rHu IFN?α on proliferation of scar FB. The effects of the above factor on DNA synthesis of scar FB were measured by means of 3 H?Thymindine incorporation ( 3H?TdR). Flow cytometry was also employed to measure the cell division cycle. Results: A decrease in cell numbers with increasing rHu IFN?α concentration showed the difference was significant (P<0 01). Correspondingly, there was a significant decrease of 3 H?TdR uptake in rHu IFN?α treated group (P<0 01) Although, the proportion of scar Fbs in stage S of cell cycle was much lower after 24 h, the change disappeared by 72 h. Conclusions: It can be concluded that rHu IFN?α could inhibit scar Fbs proliferation and their synthesis of DNA. The mechanism of rHu IFN?α inhibiting scar Fbs proliferation inhibits to go into the stage S of cell generation cycle and this inhibiting effect is of time dependent?manner.
