摘要
目的:初步测定人小静脉平滑肌细胞核DNA含量的正常参考值,确定常温下断指再植时限。方法:22例断指病人:常温缺血时间为3~10h,年龄27~54岁;分别于近、远端取小静脉组织标本,4%福尔马林固定24h,常规石蜡切片、Feulgen染色、自动图像分析仪测量细胞核DNA含量。结果:常温下人小静脉平滑肌细胞核DNA含量在缺血10h内无明显下降,所接手指全部成活。结论:小静脉平滑肌细胞核DNA含量为27.18~32.08时有断指再植指征。
Objective: To preliminily define the normal value of DNA content in nuclei of smooth muscle in humen venule. Methods: 22 patients of digit complete amputations were replanted by microsurgical technique. The warm ischemic times ranged from 3-10 hours, and ages ranged from 27-54 years. Venule specimen were acquired at proximal extremities( normal group) and distal extremities(ischemic group), and fixed in 4% formalin for 24 hours. After paraffin sections were made, Feulgen reaction was taken. Then the sections were measured by image cytometroy(ICM). Results: the DNA degradation in nuclei of smooth muscle in humen venule was not significantly if ischemic time limited in 10h, all of the replanted digits sur-vived.Conclusion: The DNA content in nuclei of smooth muscle in humen venule should be 27.18-32.08. Under this condi-tion, severd digits should be replanted.
