摘要
目的:观察17-β雌激素对过氧化氢(H_2O_2)所致PC12细胞毒性作用的影响。方法:PC12细胞暴露于不同浓度的H_2O_2或H_2O_2加17-β雌激素,观察PC12细胞的细胞计数,MTT代谢率与LDH释放率。结果:H_2O_2引起剂量依赖性的细胞计数及MTT代谢率减少,LDH释放率升高;而H_2O_2加17-β雌激素引起细胞计数及MTT代谢率升高,LDH释放率减少。结论:17-β雌激素具有对抗H_2O_2所致PC12细胞毒性的作用。
Objective: To study the effects of 17β- estradiol on hydrogen peroxide(H2O2) - induced PC12 cells cytotoxicity. Methkds: PC12 celis were exposed to H2O2 in various concentrations or treated with 17β- estradiol before exposure to H2O2, the cell count, MTT metabolic rate and LDH Ieakage rate were used to measure the viability of PC12 celis.Results: A dose-dependent decrease in cell count and MTT metabolic rate, a dose - dependent increase in LDH Ieakage rate were found in the PC12 celis exposed to H2O2- An elevation of cell count and MTT metabolic rate, a decrease in LDH Ieakage rate were revealed in the PC12 celis treatment with 17β- estradiol before exposure to H2O2.Conclusion: 17β- estradiol may be useful to reduce H2O2 - induced PC12 celis cytotoxicity.
