摘要
目的 :建立以波长为 4 2 0~ 4 6 0nm的蓝光预先照射高浓度胆红素血清样本一定时间后再测定血清尿酸的新方法 ,以消除血清高浓度胆红素对尿酸酶 过氧化物酶偶联Trinder反应法测定尿酸的负干扰。方法 :使用2 0W蓝光灯管对含不同浓度胆红素的血清样本进行一定时间的照射后 ,在全自动生化分析仪上测定尿酸浓度 ,对比光照前后的变化 ,并进行重复性实验和回收实验。结果 :蓝光照射后的血清样本胆红素浓度明显下降 ,尿酸浓度明显上升 ,重复性实验结果批内CV为 2 7% ,批间为 7 6 %。回收实验的回收率原法为 5 5 9%~ 70 5 % ,新方法为89 8%~ 113 4 %。结论 :蓝光照射法可有效消除血清尿酸测定中胆红素负干扰 ,其方法简单 ,结果准确可靠。
Objective: To establish a new method to minimize bilirubin interference on determination of uric acid by the uricase-peroxidase coupled reaction in sera. Methods:Sera samples with different bilirubin levels were pre-irridiated by blue light at 420-460 nm for a period of time before determination of uric acid. Determination of uric acid was carried out by clinical chemistry analyzer and was compared with that of the control group without being pre-irradiated by blue light. Furthermore, repetition test and recovery test of the novel method was done. Results:The concentration of bilirubin in sera after being irradiated by blue light was decreased significantly together with concentration of uric acid increased remarkably. The CV of intra-group was 2.7 % and the CV of inter-group was 7.6 % . The rate of recovery was 89.8 %-113.4 %, while that of the old method was 55.9 %- 70.5 %. Conclusion:The blue light irradiation method can minimize bilirubin interference on determination of uric acid by uricase-peroxidase coupled reaction. The method is simple and cheap. The result is accurate and reliable. It is an ideal method to diminish bilirubin interference in some assays.
