中药三七对脑缺血再灌注损伤后神经可塑性的影响 被引量:19

Effect of Chinese Traditional Medicine Sanqi on Neuronal Plasticity after Focal Cerebral Ischemia Reperfusion Injury

摘要 目的 :探讨中药三七对局灶性脑缺血后神经再塑和功能恢复的影响。方法 :5 1只Wistar大鼠分为三七组、对照组和假手术组 ,采用线栓法阻断大脑中动脉制作局灶性脑缺血模型 ,缺血 1h后恢复再灌注 ,同时三七组经腹腔注射中药三七。在术后 6h ,1、3和 7d 4个时间点应用免疫组织化学方法检测 3组缺血灶周围皮质区和海马区神经可塑性分子标志物生长相关蛋白GAP 4 3和微管相关蛋白MAP 2表达的变化 ,并于再灌注后 6h评定肢体功能恢复情况。结果 :缺血后大鼠出现左前肢瘫痪 ,三七组恢复情况优于对照组 ;再灌注后 6h ,1、3和 7d时GAP 4 3表达水平逐渐上升 ,三七组高于对照组 (P <0 .0 5 )。缺血再灌注后MAP 2表达水平降低 ,于再灌注 1、3和 7d上升 ,三七组在相应时间点高于对照组 (P <0 .0 5 ) ,但低于假手术组。结论 :局灶性脑缺血后在缺血周围区神经元出现结构重塑 ,中药三七治疗可加速这一过程并促进功能恢复。 Objective: To investigate the effects of Sanqi on neuronal plasticity and functional recovery after focal cerebral ischemia. Methods:Using suture occlusion technique, the right middle cerebral artery in Wistar rats was occluded for 1 h. Then the suture was removed for reperfusion. The animals were randomly divided into Sanqi-treated group ( n =12) and control group ( n =12). The rats in the Sanqi-treated group were subjected to the intraperitoneal infusion of Sanqi (10 mg/kg), while those in the control group were subjected to intraperitoneal infusion of normal saline. After 6 h, 1 day, 3 days and 7 days of reperfusion the animals were decapitated and brains were removed and embedded onto slides with paraffin. Immunohistochemistry was carried out to investigate the changes of neuronal plasticity markers GAP43 and MAP2. Results:The level of GAP43 expression was increased gradually while the MAP2 was decreased at the time point of 6 h and then increased gradually. The expression levels of the two proteins in the Sanqi-treated group were higher than in the control group ( P <0.05). Conclusion:Sanqi treatment can promote the neuronal plasticity following focal cerebral ischemia.
