肝毛细线虫虫卵可溶性抗原诱导大鼠保护性免疫的研究 被引量:1

摘要 目的 探讨肝毛细线虫虫卵可溶性抗原 (SEA)诱导保护性免疫的效果。 方法  30只 SD大鼠随机分为 、 、 3组 ,每组 10只。 组和 组分别予以 0 .1ml4 0 0 μg/ 0 .1ml和 2 0 0 μg/ 0 .1ml SEA加等量的弗氏完全佐剂 (FCA)每周腹腔注射免疫大鼠 1次 ; 组为对照组 ,每周腹腔注射 0 .1ml生理盐水加等量的 FCA,共 3次。各组末次免疫 1周后 ,用约5 0 0粒 /鼠肝毛细线虫感染期虫卵感染大鼠 ,攻击感染 4周后 ,剖杀大鼠检获虫卵 ,计算每克肝卵 (L EPG)及减卵率 ,并检测大鼠血清特异性抗体 Ig G和血液嗜酸性粒细胞含量。 结果  4 0 0 μg和 2 0 0 μg免疫原组减卵率分别是 5 0 .7%和 2 4 .9% ,两免疫组 L EPG、抗体水平及血液嗜酸性粒细胞含量均高于对照组 (P<0 .0 1)。 结论 肝毛细线虫 SEA可诱导大鼠产生保护性免疫。 Objective To explore protective immunity effect against Capillaria hepatica infection induced by soluble egg antigen (SEA) in rats. Methods 30 SD rats were randomly divided into Ⅰ, Ⅱ and Ⅲ groups. In group Ⅰ, each rat was immunized with 0.1 ml 400 μg/0.1 ml SEA mixed with 0.1 ml FCA; In group Ⅱ, each rat was immunized with 0.1 ml 200 μg/0.1 ml SEA mixed with 0.1 ml FCA; In group Ⅲ, each rat was immunized with 0.1 ml physiological saline mixed with 0.1 ml FCA as control. All rats in the three groups were immunized by intraperitoneal injection once a week for 3 times. A week after the final injection, all rats were challenged with 500/mouse infective stage egg. 4 weeks later, rats were killed and the LEPG were obtained and counted. The serum antibody and the eosinophils of experimental rats were carried out as well. Results In group Ⅰ and Ⅱ, the egg reduction rates were 50.7% and 24.9% respectively. The LEPG, the antibody A levels and the eosinophils were increased in the two immunized groups compared with those in control group respectively(P<0.01). Conclusion SEA of C. hepatica could induce protective against the infection of C. hepatica in rats.
