摘要
分别以潮霉素磷酸转移酶基因(hpt基因)和除草剂抗性基因(bar基因)作为筛选标记基因的载体和农杆菌组合转化水稻光温敏核不育系粳稻品种农垦58S和籼稻品种培矮64S,以两个筛选标记基因的相应筛选剂潮霉素(Hyg)和PPT(Basta)对水稻愈伤组织进行筛选研究,初步确定了不同筛选剂对不同水稻愈伤组织的筛选浓度,并比较了两个筛选标记基因的筛选效果.结果表明,不同水稻品种对筛选剂的浓度要求不一样,粳稻品种以Hyg50mg/L,PPT30mg/L较为适合;籼稻品种以Hyg50mg/L,PPT10mg/L较为适合.PPT(Basta)对粳稻品种农垦58S筛选有效,而对籼稻品种培矮64S来说则易产生假阳性.Hyg作为水稻愈伤组织的筛选剂比PPT(Basta)好.
Calli from photothermosensitive genic male sterile rice Nongken 58S (japanica) and Peiai 64S (indica) were transformed with Agrobacterium LBA 4404 harboring plasmid TAC 17 or TAC 27. Concentration of Hygromycin and PPT(Basta) were tested for two rice varieties and selection efficiencies were compared by selecting with two selection reagents. The results showed that the different rice varieties demanded different concentration of selection reagents. Hygromycin 50 mg/L,PPT 30 mg/L is efficient for Janopica, and Hygromycin 50 mg/L,PPT (Basta) 15 mg/L for Indica. PPT (Basta) is valid for Japanica variety, Nongken 58S, however, no valid for Indica variety, Peiai 64S. Selection efficiency of Hygromycin for rice transformants is better than that of PPT (Basta).
