全自动酶标仪测定淡色库蚊抗药性应用研究 被引量:3

摘要 〔目的〕根据蚊虫抗性产生的生化机制 ,利用生化检测法讨论蚊虫体内酯酶与抗性的关系 ,测定非特异性酯酶(Non -specificesterase,NSE)活性以判断其抗药性 ,探讨检测蚊虫抗药性敏感快速的检测方法。〔方法〕分别以α -乙酸萘酯和β -乙酸萘酯为底物 ,固蓝B盐为显色剂 ,用酶标板测定蚊虫NSE的活性 ,在酶标仪上读取OD值 ,同时用生物测试法检测LC50 作为对照。〔结果〕现场品系淡色库蚊NSE活力均高于敏感品系 ,与生物测试结果相一致。〔结论〕使用全自动酶标仪可以快速测定淡色库蚊体内NSE活性 ,简单易行 ,可以作为一种淡色库蚊抗药性检测方法 ,以便推广应用。 睴bjective〕 According to biochemical mechanism of insecticidal resistance in mosquitoes ,the bioassay was made to disscuse the relation between esterase and insecticidal resistance in mosquitoes, to assay NSE level and estimate the insecticidal resistance in mosquitoes and to explore a high-speed and sensitive method for the determination of insecticidal resistance in mosquitoes.〔Method〕 Taking a -Naphthyl acetate and μ-Naphthyl acetate as substrate and fast blue B salt as stain ,using automated immunoanalyser detect NSE activity of mosquitoes,the OD value is contrasted with LC50 of bioassay.〔Results〕 The NSE activity was higher in resistant strains than in susceptible strains.the result is the same as bioassay.〔Conclusion〕 The automated immunoanalyser can rapidly determine the NSE activity in mosquitoes, and easy to perform, so it can be used for quantitatively determining the resistant level of Culex pipiens pallens and convenient to be popularized.
